If your builder dies, a new male should spawn and take up the job.. might take a little while but is should happen.
You CAN have more than one of each type of worker, however it is not recommended for builders as if two builders try and build the same house it causes problems..

You shouldn't have to place a new chest at all.. If the builder dies, either an existing unemployed male will take the job, or a new male will spawn to replace the dead guy... he should then take the job.. as I say can take up to 10 minutes or so to spawn sometimes..

Also the replacements can get killed on the way sometimes. I use mo creatures and I built a city in ice plains. Polar bears and snow leopards are mean. Everytime I passed through my colony I was like No resources? No workers? WTF!!! I found without me there to guard them the villagers were getting masacred constantly and so it was a constant cycle of new colonists. Also being as the level 1 huts have grass floors mobs were spawning in their houses at night and killing them as well (I dont know the exact details of how mo creatures spawns but I do know that light level doesnt prevent animals hostile to the colonists from spawning).
Long story short, there is definitly no need to rebuild citizen chest. You just have to wait.
